В настоящее время культивированию в лабораторных условиях поддается около 1 % всех существующих бактерий. Отсутствие возможности получения чистых культур многих микроорганизмов создает серьезные трудности в фундаментальных и прикладных исследованиях. Особенно данная необходимость касается почвенных бактерий, в связи с богатым видовым разнообразием прокариот в данной природной среде. Вследствие данных фактов остро стоит вопрос поиска способов культивирования новых штаммов, в том числе разработка новых питательных сред и модификация существующих путем внесения ростовых факторов. Целью настоящей работы является изучение возможности повышения культивируемости бактерий путем внесения в среду R2A таких ростовых факторов, как гем и гумат калия, а также замены агар-агара на геллановую камедь.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Объектами исследования являлись почвы антропогенных ландшафтов Ростовской области. Было исследовано влияние ростовых факторов на культивируемость почвенных бактерий, путем подсчета общего числа колоний на универсальной питательной среде, а также отдельного учета быстрорастущих и медленнорастущих штаммов. Обработка полученных данных проводилась в программе Statistica Star Soft.</p></sec><sec><title>Результаты исследования</title><p>Результаты исследования. Согласно полученным результатам, гумат калия оказывает стимулирующее действие в низких концентрациях и ингибирующее в высоких. Гем увеличивает скорость быстрорастущих штаммов, однако одновременно с этим происходит подавление медленнорастущих бактерий. Геллановая камедь показала улучшение культивируемости медленнорастущих штаммов в одном из вариантов.</p></sec><sec><title>Обсуждение и заключение</title><p>Обсуждение и заключение. В связи с неоднозначностью результатов исследования действия геллановой камеди и гема необходимы дальнейшие эксперименты с данными компонентами. В то же время, гумат калия оказал стимулируюшее действие на рост бактерий. В связи с полученными данными можно сделать вывод, согласно которому при внесении ростовых факторов станет возможно культивировать ранее некультивируемые почвенные бактерии. Таким образом, данная тема является перспективным направлением в экспериментальной микробиологии и требует дальнейших схожих исследований.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. Currently, approximately 1% of all known bacteria can be cultivated under laboratory conditions. The inability to obtain pure cultures of many microorganisms creates significant challenges for fundamental and applied research. This is particularly true for soil bacteria because of the high diversity of prokaryotic species in this environment. These findings highlight the need to develop methods for cultivating new strains, including the development of new culture media and modification of existing media by adding growth factors. The aim of this study is to investigate the possibility of increasing bacterial cultivability by adding growth factors such as heme and potassium humate to the R2A medium, as well as replacing agar-agar with gellan gum.</p></sec><sec><title>Materials and Methods</title><p>Materials and Methods. The study involved soils from anthropogenic landscapes in the Rostov Region. The effects of growth factors on the cultivability of soil bacteria were investigated by counting the total number of colonies on a universal culture medium and separately recording fast-growing and slow-growing strains. The data obtained were processed using Statistica.</p></sec><sec><title>Research Results</title><p>Research Results. According to the results, potassium humate has a stimulating effect at low concentrations and an inhibitory effect at high concentrations. Heme increases the growth rate of fast-growing strains while simultaneously inhibiting slow-growing bacteria. Gellan gum improved the cultivability of slow-growing strains in one experimental variant.</p><p>Discussion and Conclusion. Given the ambiguous results regarding the effects of gellan gum and heme, further experiments with these components are needed. However, potassium humate stimulated bacterial growth. Based on these data, it can be concluded that the addition of growth factors may enable the cultivation of previously uncultivated soil bacteria.
